چکیده
Glutathione is an important thiol-containing compound involved in the detoxification process in erythrocytes. Its thiol group reacts with a variety of xenobiotics in a glutathione S-transferase catalyzed reaction to form conjugates that are effluxed from the erythrocytes by an ATP dependent transport mechanism. A well studied experimental system is the transport of the conjugate of glutathione and 1-chloro-2,4-dinitrobenzene. We investigated whether N-acetyl-L-cysteine protects the free-SH content and restores 1-chloro-2,4-dinitrobenzene detoxification in erythrocytes or replaces glutathione in detoxification process in glutathione predepleted erythrocytes. Our results indicate that N-acetyl-L-cysteine restores the intracellular free-SH content following depletion by 1-chloro-2,4-dinitrobenzene and N-ethylmaleimide. N-acetyl-L-cysteine (10 mM) increased the intraerythrocyte free-SH level to 14 ± 1 μmol/ml erythrocytes in 10 min in erythrocytes treated with N-ethylmaleimide. The control level was 5 ± 0.1 μmol/ml RBC. Results showed that N-acetyl-L-cysteine, in the presence and absence of L-buthioninesulfoximine, significantly recovered the 1-cloro2,4-dinitrobenzene detoxification process in erythrocytes. The rate of conjugate transport in glutathione predepleted and N-acetylL-cysteine treated erythrocytes was 449 ± 38 nmol/ml erythrocytes. In the absence of N-acetyl-L-cysteine the rate of transport was 214 ± 21 nmol/ml erythrocytes which remained similar to the control. Our results suggest that N-acetyl-L-cysteine recovers the dinitrophenyl-glutathione transport and also replaces glutathione in the detoxification of 1-cloro-2,4-dinitrobenzene in glutathione predepleted erythrocytes.

Purification and characterization of human platelet glutathione-S-transferase.
A glutathione-S-transferase was isolated and purified to homogeneity from human platelets. With a combination of ammonium sulfate fractionation and chromatographic methods, 0.2 mg of pure enzyme was obtained from 9 X 10(11) platelets with a 12% recovery. The purified enzyme had a specific activity of 7.5 U per milligram, representing an approximately 1,100-fold purification. The enzyme was foun...

The reversibility of the conjugation reaction of the diuretic drug ethacrynic acid (EA), an alpha,beta-unsaturated ketone, with glutathione and glutathione S-transferase P1-1 (GST P1-1) has been studied. When the glutathione conjugate of EA was incubated with a 5-fold molar excess of N-acetyl-L-cysteine or GST P1-1, a time-dependent transfer of EA to N-acetyl-L-cysteine or GST P1-1 was observed...

Rat liver glutathione S-transferase 3-3 (GST, EC 2.5.1.18), a triple mutant with all three cysteine residues replaced with serine (CallS) and a quadruple mutant with a Tyr-115 to phenylalanine substitution on CallS (CallSY115F) were overexpressed in Escherichia coli under the control of a phoA promoter. Using this system, we obtained over 35 mg of fully active pure protein/litre of cell medium....
متن کاملAtrazine Resistance in a Velvetleaf ( Abutilon theophrasti ) Biotype Due to Enhanced Glutathione S - Transferase Activity 1
We previously reported that a velvetleaf (Abutilon theophrasti Medic) biotype found in Maryland was resistant to atrazine because of an enhanced capacity to detoxify the herbicide via glutathione conjugation (JW Gronwald, Andersen RN, Yee C [1989J Pestk Blochem Physiol 34: 149-163). The biochemical basis for the enhanced atrazine conjugation capacity in this biotype was examined. Glutathione le...

The formation of S-nitrosoglutathione (GSNO) from amyl nitrite and n-butyl nitrite was studied in rat liver microsomes, employing N-ethylmaleimide (MalNEt) as an activator and indomethacin as an inhibitor of microsomal glutathione S-transferase (GST). Rates were compared with GST activity measured with 1-chloro-2,4-dinitrobenzene (CDNB) as a substrate. MalNEt stimulated GST activity and the for...
